Crestor and other cholesterol-lowering drugs are now widely available in the U. S. for low-to-mid-life use and for more modest dosage strengths. But a study of two years' worth of these drugs has found that their safety profile is not as strong. A study of patients at high risk of cardiovascular disease found that one-fourth of them are taking one of the drugs.
The findings, published in the journal American Heart Association, show that Crestor and other drugs are safe and effective treatments for heart disease. They are not as effective in lowering high-density lipoprotein-cholesterol (HDL-C), or in lowering triglyceride levels, which are two important risk factors for heart disease. But they are more effective in lowering levels of total cholesterol, a measure of HDL-C. The drugs have been found to have fewer side effects than their statins, although they can cause serious side effects.
In the study, researchers used data from the FDA's database of more than 5,000 patients with high-risk cholesterol who were randomly assigned to take a statin for at least six months and then monitored their lipid levels for six months or more. They found that the most common adverse effects were low-density lipoprotein (LDL-C), cholesterol levels of 30% to 40%, triglycerides of 2.6 to 2.9 mmol/L, and fasting blood sugar levels of 30%. The drugs were generally safe and well-tolerated.
But many of the drugs had more serious side effects. One of the drug's most common adverse effects was the liver. The drug was also associated with a higher risk of liver failure, and its use was associated with a small but statistically significant increase in blood pressure and cholesterol levels, especially at high doses. It also caused more weight loss and slowed the progression of the disease.
The risk of liver problems in patients taking Crestor, which is also known as atorvastatin, was increased by about 5% and 1.8% in patients taking the drugs in the same group, compared to the placebo group. Patients taking the drugs in the same group also had higher doses of the cholesterol-lowering drug, simvastatin.
These drugs were generally safe and well-tolerated. In addition to the high cholesterol and the risk of heart disease, the drug did not cause more deaths. The drug was associated with a higher risk of myopathy and other conditions that were not listed on the labels.
The study also found that the drug was less likely to cause anemia. The drug was not associated with a reduction in serum creatinine. The drug was associated with a lower rate of mortality.
The drugs were generally well-tolerated and safe. But they did have serious side effects that researchers were not able to rule out.
The results are "a landmark step forward in the treatment of heart disease," said study co-author John D. Korn, who was not involved in the study. "These drugs were safe and effective treatments for heart disease, but they had a high risk of serious side effects."
Korn said the drugs were safe and effective for patients with high-risk cholesterol and high-density lipoprotein-cholesterol (HDL-C) cholesterol levels and had a favorable safety profile.
The drugs were safe and well-tolerated, Korn said. "These drugs were effective in lowering high-density lipoprotein (HDL-C), and we saw no serious side effects."
However, Korn said, there were some serious side effects that the drugs were not associated with. "Some of the serious adverse effects that led to this study may have been caused by the drugs or side effects of the other drugs we looked at," he said. "The drug is a statin, and it is the same drug. There are other drugs in this class that are not statins and are not associated with a high risk of liver problems."
Crestor, a cholesterol-lowering drug, is currently being studied in patients with high-risk cholesterol. The drug is already in Phase III trials to try to determine its effects on the risk of heart disease, and Crestor is being tested for other potential effects on the heart.
Korn said the drugs may also be used in other patients who have high-risk cholesterol or other health problems, and the drugs are being studied to increase the risk of cardiovascular disease.
